How long will it take to get from the Times Square area to Chinatown and what is the best way to get there?

If you take the Broadway BMT subway (the N, R, Q or W line) from Times Square to Canal Street (which is what I would do), it should take you about eight minutes.

I believe the N will be slightly faster than the R or W, since it runs express in Manhattan.

The N is indeed slightly faster -- if both express and local arrive simultaneously, take the N. However, the difference over that distance is not significant enough to pass up an earlier-arriving local train in order to wait for the express.
